微學程選修學生之心得 (108學年修課學生回饋) 首先想先感謝高醫校方無償提供系上課程外,如此多元化的學分學程/微學程之課程,讓我們學生能以探索自己系所外專業領域之興趣,可以慢慢了解自己喜歡甚麼,以及對於可能適合從事甚麼領域……等的專業興趣培養。更重要的是,選修了學分學程及學分微學程,讓我們學生在大學經歷四年畢業後,能比同期學生多了更多競爭力。 我選擇選修的學分學程是「臨床試驗微學分學程」,這領域的學分學程課程與系上專業領域不盡相同,讓我自己的課業壓力有強大的挑戰,但也因此拓展開闊不同領域的知識,其中一門公衛系王姿乃教授開授的選修「流行病學」,讓我對此方面專業特別印象深刻,師長講解淺顯易懂,因應武漢肺炎疫情時事,在閱讀新聞或媒體資訊時也能看得懂些專有名詞的意義,相當實用。另外,在生物系陳炤杰教授開授的選修「試驗設計」、「試驗設計實習」也是受益良多,小班制的教學使我們每個學生能在課堂的軟體操作上即時接受老師的指導。

研究興趣:
細胞凋亡(apoptosis)是我們最熟悉的程序性細胞死亡。多細胞生物為了發育與維持恆定而啟動程序性細胞死亡機制,特徵包括細胞膜皺縮、細胞內膜磷脂質外翻(PS Flipping)、DNA斷裂及形成凋亡小體並且被周圍細胞或巨噬細胞吸收,而不會產生發炎反應。少數真菌或原生動物也有被報導有細胞凋亡產生,如黏黴菌Dictyostelium discoideum, Trypanosoma cruzi 與 Trypanosoma brucei rhodensiense , 鞭毛菌Tetrahymena。目前癌症病人主要治療方式之一為小分子藥物為基礎的化學治療,並且過去大部分的抗癌藥物設計原理主要著重於誘導細胞凋亡,希望能避免發炎產生降低對病人的副作用。然而近年來大量研究與臨床報告指出癌症細胞的凋亡路徑常常是突變缺陷或是最後對凋亡誘導的抗性,導致對化療效果不佳。自凋亡機制被發現至今,至少有12種以上的新穎程序性細胞死亡被報導,如何利用與調節這些新穎程序性細胞死亡可能成為重要癌症治療策略。
我們實驗室的主要興趣則是對於這些新穎程序性細胞死亡方式如何在癌症治療應用。1. 我們實驗室主要探索台灣常見的癌症包括肝癌、乳癌及非小細胞肺癌如何對臨床藥物產生抗性。2. 透過了解癌細胞產生抗藥性機制進行預防或發展使癌細胞對臨床藥物重新致敏化 (Chemosensitizer)的策略。3. 除了細胞凋亡 (Apoptosis),我們也對新穎非凋亡性細胞死亡(Non-apoptotic programmed cell death)包括鐵凋亡 (Ferroptosis)、程序性壞死(Necroptosis)、細胞焦亡 (Pyroptosis)及類凋亡 (Paraptosis) 有興趣,希望能運用癌細胞本身特性以誘導細胞死亡以抑制癌細胞,並且能夠透過了解這些程序性的細胞死亡機制,未來就能運用在其他疾病的治療。

苯氧酚化合物diTFPP能透過調控肝細胞癌的外源性C2-神經醯胺代謝,並調節ROS的形成和自噬壓力而誘導肝癌細胞凋亡

Antioxidants ( IF 5.014 ) DOI: 10.3390/antiox10030394 . 簡介:外源性合成短碳鏈神經醯胺可抑制癌細胞生長及誘導細胞死亡。然而細胞內醯胺代謝途徑會傾向將外源性C2-神經醯胺代謝成向存活性醯胺而導致C2-神經醯胺誘導的抗肝癌細胞作用減弱。 而苯氧酚化合物diTFPP可能協助調節細胞內鞘脂醯胺代謝,使C2-神經醯胺為促凋亡並增強ROS的產生和自噬壓力,並最終使肝細胞癌對於對C2-神經酰胺誘導凋亡致敏化。
除了癌症領域,細胞生與死的平衡失調是導致許多疾病產生主因,因此我們也探討疾病包括眼部退化預防探討薑黃素衍生物於藍光照射視網膜色素上皮層細胞的保護機制

粒線體對人體疾病影響的轉譯研究與預防策略 [與本校臨床醫學教師 林祐賢助理教授合作]
林祐賢醫師為腎臟內科主治醫師, 實驗室為針對粒線體於人體疾病所扮演角色的轉譯實驗室。粒線體是細胞能量工廠, 主要利用Krebs Cycle 產生ATP 提供細胞能量。粒線體有分內膜、外膜。藉由訊息調控, 訊息因子不單單只是存在於細胞質(cytoplasm), 更會進出粒線體, 藉由電子傳遞鍊(electron transport chain), 調控complex I-V, 影響了O2、H2O的製造。也會導致ROS的產生。現在的研究證據顯示、粒線體不單單只是扮演提供細胞能量, 更在細胞凋亡扮演重要角色。林醫師實驗室, 利用細胞、動物模式, 與加州大學爾灣分校王秉訓教授實驗室證明了粒線體AKT1訊息的調控, 在缺血再灌流(ischemia-reperfusion)急性腎損傷(acute kidney injury, AKI)會導致腎臟細胞的損傷與修復。本研究獲得Kidney International (impact factor=8.95, ranking 6/85, Q1) (Kidney Int. 2021 Apr;99(4):870-884).
Translational Research of Mitochondrial Metabolism in Human Diseases [Collaboration with Dr. Hugo Y.-H. Lin, MD, Ph.D. of KMU Hospital]
Dr. Lin is a physician-scientist of the Division of Nephrology, and his laboratory is focusing on translation research of the role of mitochondria in human diseases. Mitochondria are cellular energy factories, which mainly utilize the Krebs Cycle to generate ATP for providing cellular energy. Mitochondria have inner and outer membranes. Through the signal’s transduction, the molecular signal not only exists in the cytoplasm but also translocates into the mitochondria. Through the electron transport chain, it regulates complex I-V and affects the production of O2 and H2O. It can also lead to the generation of ROS. Current research evidence shows that mitochondria not only play a role in providing cellular energy but also play an important role in apoptosis. This study was awarded by Kidney International (impact factor=8.95, ranking 6/85, Q1)

In summary, the cellular growth curves for two CNS tumors (C6 and Y79) were established by evaluating DT's population. This study concludes that the cell line’s growth curve pattern is dependent on the seeding density. The growth curves constructed from 4,000 cells/cm2 show a more expected growth curve pattern, including the lag phase, exponential phase, plateau phase, and death phase. Increasing the seeding density changes some properties of the cells. These changes are cell line-specific. It is thus suggested that researchers consider the effect of seeding density before running any experiments. This study also helps decide and choose the best cell number and does well with prior knowledge about this cell line.

一般而言,酸性胞器所需的pH梯度會被酒精或醛改變,因此LysoTracker 大多情況僅可用於活細胞,而不能用於固定組織。 但有些時候可以先用lysotracker標定溶小體,但一定要在固定液使用之前。
另外,也可以以用溶酶體標記物LAMP1 / LAMP2 /組織蛋白酶B來標記溶酶體。
Since the pH gradient needed to stain acidic compartments is altered by alcohol or aldehydes, LysoTracker can only be used in live cells and cannot be used in fixed tissues. Stains may be used for this, but only after they've been lysosome stained. Alternatively, several lysosomal markers LAMP1 / LAMP2/ cathepsin B, which can be stained with LC3, can be used to label lysosomes.
